Pollen wall development in Sorghum bicolor is morphologically and temporally paralleled by the formation of a prominent orbicular wall on the inner tangential surface of the tapetum. In the late tetrad stage, a thin, nearly uniform primexine forms around each microspore (except at the pore site) beneath the intact callose; concurrently, small spherical bodies (pro-orbicules) appear between the undulate tapetal plasmalemma and the disappearing tapetal primary wall. Within the primexine, differentially staining loci appear, which only develop into young bacula as the callose disappears. Thus, microspore walls are devoid of a visible exine pattern when released from tetrads. Afterwards, sporopollenin accumulates simultaneously on the primexine and bacula, forming the exine, and on the pro-orbicules, forming orbicules. Channels develop in the tectum and nexine, and both layers thicken to complete the microspore exine. Channeled sporopollenin also accumulates on the orbicules. A prominent sporopollenin reticulum interconnects the individual orbicules to produce an orbicular wall; this wall persists even after the tapetal protoplasts degenerate and after anthesis. While the pollen grains become engorged with reserves, a thick intine, containing conspicuous cytoplasmic channels, forms beneath the exine. Fibrous material collects beneath the orbicular wall. The parallel development and morphological similarities between the tapetal and pollen walls are discussed.  相似文献

We have isolated a new class of deletion mutants of phage lambda that extend from the prophage attachment site, att, into the gam and cIII genes. In this respect they are similar to certain of the λpbio transducing phage, but they differ in having a low burst size and in forming minute plaques. Lytically grown stocks of the deletions contain a variable proportion of phage that produce large plaques. These have been shown to carry an additional point mutation. Similar mutations, called chi, have been described by Lam et al. (1974), who showed that they result in a hot-spot for recombination produced by the host recombination system (Rec). We show that chi mutations can occur at several sites in the lambda genome and produce a Rec-dependent increase in the burst size of the one deletion tested.—In addition to reducing burst size, the one deletion tested reduces synthesis of DNA and endolysin but increases production of serum blocking protein. A chi mutation partially restores DNA synthesis and endolysin production and reduces serum blocking protein to normal levels. Our results are consistent with the hypothesis put forward by Lam et al., that chi enhances the frequency of Rec-promoted recombination, which provides the only pathway for production of maturable DNA in a red- gam- infection. The mechanism of the differential effect on protein production is, however, unclear.—Chi mutations are found to occur in DNA other than that of λ. We show that, as has been suggested elsewhere (McMilin, Stahl and Stahl 1974), the λpbio transducing phages carry a chi mutation within the E. coli DNA substitution. A chi mutation also arose in a new substitution of unknown origin isolated in the course of this work.  相似文献

The presence of high concentrations of sulfate, iron, and hydrogen (acid) ions in drainage from coal mines and other areas containing waste pyritic materials is a serious water pollution problem. Sulfate can be removed from solution by microbial reduction to sulfide and subsequent precipitation as FeS. A mixed culture of microorganisms degraded wood dust cellulose, and the degradation products served as carbon and energy sources for sulfate-reducing bacteria. Metabolism of carbon compounds resulted in a net pH increase in the system. Oxidation-reduction potential (Eh) and temperature and carbon supplements were studied in an effort to accelerate the sulfate reduction process, with the ultimate objective of utilizing the process as a pollution abatement procedure.  相似文献

An enzyme-linked immunosorbent assay for 6-keto prostaglandin F, a stable metabolite of prostacyclin, has been developed. The assay allows quantitation of 6-keto PGF in the range 1–200 pg/0.1 ml and shows very low cross reactivity to nine other prostaglandins. Dose dependent stimulation by thrombin of 6-keto PGF formation in human endothelial cells in culture has been used to verify the assay. Quantitation by the enzyme linked immunosorbent assay agrees closely with determination by radioimmunoassay.  相似文献

A number of copper(II) complexes of tridentate ligands with various donor atoms have been studied in an attempt to duplicate the unusual reactivity patterns and accompanying spectral changes of the copper(II) center in galactose oxidase. Results indicate that in order to match the optical and electron spin resonance spectral change observed upon CN? binding by the enzyme, an equatorial, negative ligand must be displaced in a small molecule model. The crystal and molecular structure of the best model complex was solved by a single crystal X-ray diffraction study. The compound, monoacetato-1,3-bis(2-(4-methyl-pyridyl)imino)isoindolatocopper(II), crystallizes in the centro-symmetric triclinic space group Pī with a = 7.392(3) Å, b = 13.782(5) Å, c = 23.422(12) Å, α = 92.08(3)°, β = 104.11(5)°, γ = 109.98(4)°, V = 2156(1) Å3, d(obsd.)(calc.)=(1.43)(1.44) g/cm?3 for mol wt of 466.7 and Z = 4. Diffraction data were collected with a Syntex Pl diffractometer using graphite-monochromatized Cu radiation (λ = 1.5418 Å). The copper atoms were located from a Patterson synthesis; all other nonhydrogen atoms were located via difference. Fourier techniques, and hydrogen atoms were placed in calculated positions. Final refinement resulted in discrepancy indices of R = 0.089 and “Goodness to Fit” = 3.68 for all 3608 reflections having (I) ? 3σ(I) (5°<2θ<100°). There are two unique molecules in the asymmetric unit that are monomeric and well separated. The geometry around the copper atom is approximately square pyramidal, with the coordination sphere derived from three nitrogens of the tridentate ligand, one oxygen from the acetate unit, and an oxygen atom of a water molecule occupying an axial position. The structure is surprising both in that an axial water molecule is present and that the remaining four ligand atoms to the copper atom are rather distorted from a planar configuration. The plane defined by the copper, N5, and N3 atoms intersects the plane defined by the copper, Nl, and Ol, atoms forming a “twist angle” of 25.0° (0.0° would be ideal for a planar inner coordination sphere). The stereoelectronics of the inner coordination spheres of the type II Cu(II) enzymes galactose oxidase and superoxide dismutase are discussed and appropriate comparisons are made with emphasis on the origin of spectral changes observed upon anion binding.  相似文献

Proteinase levels in the posterior midgut of fifth-instar and adult Rhodnius prolixus follow a cyclic pattern after ingestion of the bloodmeal. In the fifth instar, cathepsin B showed two peaks: the first occurred 6 days after ingestion and the second at the time of ecdysis. Cathepsin D, cathepsin B and lysosomal carboxypeptidase B reached maximal levels 6 days after ingestion. At this time the highest levels of these proteinases were found in mated females, the lowest in males and intermediate levels in virgin females. Maximal levels of aminopeptidase occurred later than catheptic enzymes, and the decline, after maximal levels were achieved, was much more gradual.Catheptic-proteinase levels within the posterior midgut in fifth-instar larvae and adults correlated positively with the amount of protein contained in this gut region. This indicates that production of these proteinases is controlled by a secretagogue mechanism. Aminopeptidase levels were controlled in a different manner. The mated state or sex of adults altered the proteinase levels by changing the amount of protein that was passed into the digestive midgut from the crop.  相似文献

The pattern of unconjugated pterins in liver tissue and in urine from patients with atypical forms of phenylketonuria with hyperphenylalaninemia (HPA) has been investigated with a high performance liquid chromatographic technique. Two patients with defects in the biosynthesis of biopterin have been shown to have higher than normal levels of neopterin and lower than normal levels of biopterin. In contrast, a patient with HPA due to a deficiency of dihydropteridine reductase has the reverse urinary pattern, i.e., high biopterin, low neopterin. These results indicate that the ratio of neopterin to biopterin in urine can be of value in discriminating between HPA due to a deficiency of phenylalanine hydroxylase (classic PKU), HPA due to dihydropteridine reductase deficiency, and HPA due to a block in the biosynthesis of biopterin.  相似文献
